Dark quencher for pFAST: switch pFAST fluorescence off in seconds, without washing, and back on at will.
TFDarth (HBIR-3M) is a non-fluorescent chromophore that forms a very tight, dark assembly with pFAST. Added in excess, it outcompetes the fluorogen bound to pFAST and turns its fluorescence off within seconds in living cells, with no wash step and no photobleaching. Adding an excess of any pFAST fluorogen switches the fluorescence back on.

Applications
Fast, reversible switching-off of pFAST fluorescence in cells and multicellular samples, sequential multicolor protocols on the same pFAST-tagged protein, and dark-acceptor designs for FRET read out by fluorescence lifetime (FLIM-FRET). It works wherever pFAST does, including anaerobic conditions.
